What is confocal microscope?
Confocal microscopes feature superior resolution, much higher than that of conventional optical microscopes, and are often used in measurement applications.
Principle of confocal optics
Confocal optics can create a high-resolution high-contrast image that is free from the interference of undesirable scattered light because it collects information from areas in focus only.
Optical sectioning effect
Confocal optics captures reflected light from the focal plane only. If you observe a three-dimensional sample using confocal optics, you will see highlighted areas at an equal height. An image you see looks like a contour cut out from the sample at the depth of focus. This effect is called optical sectioning.
